Q: Should the LRW react with LAL?

A: No, the LRW should not react with the LAL.

Q: How much LRW is required to dissolve the PYROSTAR™ ES-F Series and the Limulus Color KY series?

A: The amount of LRW depends on the testing format of the product.

  • PYROSTAR™ single test: 0.2 mL of LRW for the negative control only. The sample or dilutions (0.2 mL) that are being examined should be added to the single test lyophilized vial to reconstitute the reagent.
  • PYROSTAR™ multi test: 2.0 mL of LRW or 5.2 mL of LRW should be added to the vial, depending on the volume listed on the label.
  • PYROSTAR™ ES-F/Plate: Only 2.0 mL of LRW should be added according to the volume listed on the label.
  • Limulus Color KY single test: 0.2 mL of LRW for the negative control only. The sample or dilutions (0.2 mL) that are being examined should be added to the single test lyophilized vial to reconstitute the reagent.
  • Limulus Color KY multi test: Only 2.0 mL of LRW should be added according to the volume listed on the label.

A: The easiest way to determine the volume of LRW needed to reconstitute the CSE is to check the specific LAL lot’s corresponding Certificate of Analysis (COA). For PYROSTAR™ ES-F Series and Limulus Color KY series, a COA is provided or can be accessed at www.wakopyrostar.com.

When testing with Limulus Color KY series, the potency is shown on the CSE vial label. Divide the potency (EU/vial) on the label by 1,000 to determine the volume (mL) of LRW necessary for reconstituting the CSE to yield 1,000 EU/mL.
