Toxinometer® ET-6000 is the new equipment developed by the company Wako for the reading of tubes in which the LAL (Limulus Amebocyte Lysate) test is performed for the quantification of bacterial endotoxins in a sample. Today the LAL test is considered to be the most effective method for the detection of pyrogens since bacterial endotoxins are the most commonly found pyrogens in samples of medicine and food, among others.
The users will find a highly flexible system in Toxinometer® ET-6000, with full computer control. This equipment can be used when you want to quantify endotoxins in small or larger volume samples, easily adaptable with expansion modules when the number of samples the researcher needs to work with is very large.
Toxinometer® ET-6000 works by measuring the proportion of light passing through the tube in time, with respect to the intensity of light passing at the time of starting the test. The reaction of the amebocytes of the horseshoe crab (Limulus polyphemus) in the presence of bacterial endotoxins leads to the coagulation of proteins that act as a defense mechanism of the immune system of the crab. The appearance of the clots generates turbidity in the sample under study and thereby changes the intensity of light passing through the sample. Therefore, the operating principle of the toxinometer is very simple. The time it takes for turbidity to appear in the mixture in the tube is proportional to the amount of endotoxins present in the sample or standard being measured. At a higher concentration of endotoxins (lipopolysaccharides) in a sample, the amebocyte lysate reaction occurs faster. In this test, the time necessary for the sample of endotoxins to react with the mixture of LAL reagents is called the reaction time.
Through the measurement of the reaction time, ET-6000 can be used to find out the concentration of endotoxins in a sample. If the turbidimetric test is used, the speed with which turbidity appears is measured to calculate the reaction time. As for the chromogenic test, the increase in color intensity is measured to calculate the reaction time.
