The Use of Toxinometer ET-6000 in LAL test

The Use of Toxinometer ET-6000 in LAL testToxinometer® ET-6000 is the new equipment developed by the company Wako for the reading of tubes in which the LAL (Limulus Amebocyte Lysate) test is performed for the quantification of bacterial endotoxins in a sample. Today the LAL test is considered to be the most effective method for the detection of pyrogens since bacterial endotoxins are the most commonly found pyrogens in samples of medicine and food, among others.

The users will find a highly flexible system in Toxinometer® ET-6000, with full computer control. This equipment can be used when you want to quantify endotoxins in small or larger volume samples, easily adaptable with expansion modules when the number of samples the researcher needs to work with is very large.

Toxinometer® ET-6000 works by measuring the proportion of light passing through the tube in time, with respect to the intensity of light passing at the time of starting the test. The reaction of the amebocytes of the horseshoe crab (Limulus polyphemus) in the presence of bacterial endotoxins leads to the coagulation of proteins that act as a defense mechanism of the immune system of the crab. The appearance of the clots generates turbidity in the sample under study and thereby changes the intensity of light passing through the sample. Therefore, the operating principle of the toxinometer is very simple. The time it takes for turbidity to appear in the mixture in the tube is proportional to the amount of endotoxins present in the sample or standard being measured. At a higher concentration of endotoxins (lipopolysaccharides) in a sample, the amebocyte lysate reaction occurs faster. In this test, the time necessary for the sample of endotoxins to react with the mixture of LAL reagents is called the reaction time.

Through the measurement of the reaction time, ET-6000 can be used to find out the concentration of endotoxins in a sample. If the turbidimetric test is used, the speed with which turbidity appears is measured to calculate the reaction time. As for the chromogenic test, the increase in color intensity is measured to calculate the reaction time.

The Toximaster® QC7 software comes with the Toxinometer ET-6000 and it is the tool that enables us to achieve an efficient work routine and obtain high-quality data analysis. When purchasing the equipment, the software already comes installed so that it can be used immediately after purchase and it does not delay the immediate use of the equipment. In addition, you can find a compilation of software in the documentation that comes with the toxinometer.

The Advantages of Using Toxinometer ET-6000:

  • It is compatible with any of the methods used to perform the LAL test; the kinetic turbidimetric method, the chromogenic-kinetic method and the gel-clot method. Printable copies of the data obtained in measurements are generated, which facilitates the analysis of these data.
  • In a simple module, up to 16 samples can be measured simultaneously. To measure more samples, there are expansion modules available. With the use of these modules, 16 samples more are added for each module attached. There can be up to 8 modules, which will allow for the reading of 128 samples.
  • Measurements can be made both at 30°C and at 37°C, with a high sensitivity of 0.001 EU/mL in about 40 minutes.
  • The determination of endotoxins performed by using ET-6000 is in compliance with the American, European and Japanese Pharmacopoeia standards for this type of test, as well as FDA standards.
  • Toximaster QC7 is software which integrates the most advanced knowledge in this type of product and which facilitates the use of the equipment, allowing us to save time and achieve an early detection of any equipment failures.
